Let’s understand these key molecules, phospholipids. Phospholipid is a general name for a fat with a phosphate part. They are basic components of all cell membranes. This includes cell membranes in brain tissue structures. There are two main types of phospholipids. Glycerophospholipids (GPL) are one type. Sphingolipids are the other. A GPL molecule has a glycerol center. It is a three – carbon structure with three connection sites. A phosphate group connects to one site on the glycerol. The fatty acid tails hate water, though. They are the hydrophobic tail parts. This dual nature makes them behave in water. In water, GPLs form two layers by themselves. The water – loving heads face outward.

The water – hating tails point inward. This amazing ability forms cell membranes. They do this together with sphingolipids and proteins. Different GPLs are not distributed the same way in the membrane. They cluster in certain layers.

PC is mainly in the outer layer, for instance, often together with sphingomyelin. PE is mostly in the inner layer. PS is only found inside the inner layer. This difference in their locations matters for how cells work.

Besides consuming GPLs, our bodies make them too. PC, PE, and PS can be produced inside. Some production is from scratch, which is de novo. Other ways involve changing GPLs that are already present. That is the remodeling of molecules. The production of PC and PE starts by forming phosphatidic acid. This acid is then modified by enzymes. It becomes either 1,2 – diacylglycerol or CDP – diacylglycerol. These are starting materials for different GPLs. 1,2 – diacylglycerol helps make PC and PE. CDP – diacylglycerol is used to make phosphatidylinositol. The production of PS in our bodies does not require phosphatidic acid first. The main ways GPLs are made are complicated, but they show how GPLs are always being produced inside us.

Looking closer at how PC is made, our body mainly uses two major ways. The CDP – choline pathway builds new PC. This is a de novo pathway. The PEMT pathway remodels existing PE molecules. PE is made like PC using CDP – ethanolamine. But it can also be formed by changing PS. This happens through the PSD pathway. PS making in our cells involves exchange reactions. Special enzymes called synthases catalyze these reactions. The PSS1 enzyme exchanges free serine for choline from PC. This makes PS from PC. The PSS2 enzyme exchanges ethanolamine from PE with serine. That also makes PS. Knowing these different routes helps show how dynamic GPL making is.

Using different making ways has one interesting result. It looks like the way used changes what fats are in the GPL. PC made via CDP – choline usually has straight or one – or two – bend fatty acids. PC made via PEMT may also include many – bend fatty acids, like the omega – 3 DHA type. Some think PEMT prefers PE with omega – 3 in it. Others guess it depends on how much DHA – containing PE is around to be changed. Similar fat makeup differences are seen for PE. PE from the CDP – ethanolamine pathway mainly has one – or two – bend fats at one connection site. PE from the PSD pathway tends to contain many – bend fats, like omega – 6 ARA, or omega – 3 DHA or EPA. What the enzyme prefers also shows in PS making. Studies show that PSS1 and PSS2 prefer PC or PE with omega – 3s, especially DHA, compared to GPLs with other fats. This hints that our body’s making process can intentionally put good fats into GPLs.

We also get GPLs directly from food. People may eat about two to eight grams daily. This is about one to ten percent of daily fat intake. GPLs are in many foods we eat. Good sources include dairy products, eggs, and animal meat. Meat from cows or pigs is included, especially muscle and organ meats like brain. Fish, mainly ocean types like krill or squid, also contain GPLs. Plant foods are also sources of them. Certain seeds and beans are options, such as rapeseeds, sunflower seeds, and soybeans.

What fats are in GPLs vary greatly depending on the food source. GPLs from ocean fish like krill often contain omega – 3 fats. So do mammal brain tissues. Or eggs from hens fed omega – 3 – rich food. Soybeans have omega – 3 called ALA. Our bodies can convert ALA into longer omega – 3s, like EPA and DHA. Soybeans, fish, brain, and eggs also provide omega – 6 fats and other kinds of fats.
